4-laboratoriya mashg‘uloti.
Mavzu: Amilaza fermentining kraxmalga ta'siri.
Maqsad: Amilaza fermentining murakkab uglevod-kraxmalga ta'sirini kuzatish.
Jihozlar: Probirka, kolba, tomizgich, polipropilen o‘lchov idishlari to‘plami, distillangan suv, reaktivlar to‘plami (1% yod eritmasi, 0,5% kraxmal eritmasi), so‘lak fermenti.
Ishni bajarish tartibi:
1. Har bir o‘quvchi stoliga 2 tadan probirka tarqatiladi.
2. Uning bittasiga 2-3 ml kraxmalning 0,5% li eritmasi quyiladi
3. Ikkinchi probirkaga 2-3 ml kraxmalning 0,5% li eritmasi uning ustiga 1 ml amilaza ferment shirasi quyiladi. Vaqtni belgilab, probirkadagi aralashma chayqatiladi.
4. So‘ngra birinchi probirkaga bir tomchi yod tomiziladi. Bunda probirkadagi suyuqlik ko‘k rangga kiradi - suyuqlik tarkibidagi kraxmal yod ta'sirida ko‘k rangga kirganligini ko‘rsatadi.
5. Ikkinchi probirkaga bir tomchi yod tomizilganda amilaza fermenti shirasi qo‘shilgan probirkada uning miqdoriga qarab binafsha, qizil, sariq rang hosil qiladi.Bu amilaza ta'sirida kraxmalning parchalanishidan dalolat beradi.
