Asboblar va reaktivlar: Tarozi, termostat, 50 ml li kolbalar, byuretkalar,
100 ml li tagi tekis kolbalar, pipetkalar, voronkalar, filtr qog’oz, chinni xovoncha, maydalangan shisha, shisha tayoqcha, 1%-li vodorod peroksid; 0,1 H-li KMnO 4 , 0,1 M fosfat buferi (pH 6,8), 2 H-li sulfat kislota. Materiallar: barg, unayotgan urug’. Ishning bajarilishi: 5 g o’simlik bargidan yoki unayotgan urug’dan olib, 5 ml fosfat buferi va maydalangan shishadan solib, tisha xovonchada yaxshilab maydalanadi. Maydalangan massa 50 ml li kolbaga solinadi va fosfat bufer bilan 50 ml gacha yetkaziladi. Aralashma yaxshilab aralashtiriladi va 37°S li termostatga 15 minutga qo’yiladi va undan olib, aralashma filtrlanadi. Shundan so’ng 2 ta tagi tekis kolba olib, har biriga 20 ml dan filtratdan solinadi. Birinchi kolbaga (kontrol) 5 ml 2 H-li H 2 S0 4 dan fermentni aktivligini yo’qotishi uchun solinadi. Keyin har ikkala kolbaga 20 ml dan suv va 3 ml dan 1% li H 2 0 2 eritmasidan solinadi va 37°S li termostatda 15 minut ushlanadi. Vaqt o’tgandan keyin 2-tajriba kolbasiga 5 ml 2 H li H 2 SO 4 solinadi, aralashtiriladi va kaliy permanganatning 0,1 M li eritmasi yordamida titrlanadi. Bunda och pushti rang hosil bo’lishi va 30 sekund davomida yo’qolmasligi kerak. Katalazaning aktivligi quyidagi formula orqali hisoblanadi: V (B-A) KC nC i t bu yerda A va B - titrlash uchun ketgan kaliy permanganatning miqdori, ml da, (kontrol va tajriba uchun ketgan miqdor). K - 0,1 M kaliy permanganatni titrlash uchun tuzatish ko’rsatgichi. C - aralashma hajmi, ml da. C i - aniqlash uchun olingan aralashmani hajmi, ml da. n - o’simlik materialining og’irligi, g da. t- vaqt, soatda.
